Moderna (NASDAQ: MRNA) has seen its market capitalization surge by approximately $28 billion over the past week, driven by a landmark Phase 3 clinical trial success for its personalized melanoma vaccine. The stock closed Thursday at $133.32, up 110.5% from its August 14 close, even after a 23.55% pullback from Wednesday's peak.

Weekly Price Action

  • Aug. 14 close: $63.32
  • Aug. 19 close: $174.38 (one-day gain of 176.97%, adding ~$44.5B)
  • Aug. 20 close: $133.32 (one-day drop of 23.55%, wiping ~$16.4B)
  • Net weekly change: +110.5%, +$27.9B

The catalyst was the announcement from Moderna and Merck (NYSE: MRK) that their Phase 3 INTerpath-001 trial met its primary endpoints for recurrence-free survival and distant-metastasis-free survival. The study involved 1,137 patients with fully resected stage IIB-IV melanoma. Notably, no new safety concerns were reported.

Market Valuation vs. Sales Forecasts

Barclays analysts, as quoted by Reuters, project annual melanoma sales could reach up to $3 billion by 2035. Given the 50/50 profit-sharing arrangement with Merck, the $27.9 billion weekly market value increase equates to 9.3 times that projected annual revenue, before accounting for time value, costs, or Merck's share. This suggests investors are pricing in success beyond melanoma, including ongoing trials in lung, bladder, and kidney cancers.

Moderna's total market value now stands at $53.23 billion, more than seven times its cash and investments of $6.9 billion as of June 30. The company reported second-quarter revenue of $145 million and a net loss of $782 million, highlighting the need for continued pipeline financing.

Analyst Skepticism

Despite the surge, Wall Street remains cautious. The 15-analyst consensus rating is 2 buys, 12 holds, and 1 sell, with an average price target of $82.62—38% below Thursday's closing price. Targets range from $40 to $170, reflecting the binary nature of clinical outcomes.

Key risks include underwhelming full Phase 3 results, regulatory delays, manufacturing challenges, and slower-than-expected melanoma demand. The previous Phase 2b trial (KEYNOTE-942) showed a 49% reduction in recurrence or death risk at five years, but that was based on only 157 patients.

Looking Ahead

Investors will be watching for detailed efficacy data, including hazard ratios and survival curves, which were not disclosed. Moderna plans to share the data with regulators and continue follow-up for overall survival. The company's ability to replicate this success across multiple tumor types will be crucial to sustaining the revaluation.
