The late price implies a preliminary equity value of about $33.65 billion, based on 151.0 million shares outstanding as of July 24.
Illumina will replace Builders FirstSource, Inc. (NYSE: BLDR) in the S&P 500, with the change effective before trading opens on September 21. This marks a return to the large-cap index after a two-year absence. In June 2024, GoDaddy Inc. (NYSE: GDDY) had replaced Illumina, which then moved to the S&P MidCap 400 as the company prepared to spin off its Grail unit.
The index inclusion is expected to trigger mechanical buying from funds that track the S&P 500. However, S&P did not disclose the final index weight or the number of shares these funds may need to purchase. The value estimates remain preliminary, as employee stock issuance or buybacks could have altered the share count.
Fundamentally, Illumina's latest quarterly results provide a solid backdrop for the promotion. Revenue rose 9.5% year-over-year to $1.159 billion, while GAAP operating margin improved to 21.1% from 20.2% a year earlier. However, cash conversion weakened: free cash flow fell to $162 million from $204 million, and operating cash flow declined to $201 million from $234 million.
Management raised its 2026 revenue guidance to a range of $4.60 billion to $4.64 billion and lifted non-GAAP earnings per share guidance to $5.30–$5.40. CEO Jacob Thaysen highlighted robust demand for the NovaSeq X platform, tying it to broader workflows and multiomics capabilities.
Despite the positive signals, valuation remains a concern. At the after-hours price of $222.88, the stock trades at approximately 41.7 times the midpoint of the adjusted earnings outlook, and it stands 34.3% above the $166 average analyst price target from Benzinga's 19-analyst panel. That average includes targets set before the recent rally, and views remain mixed: 11 Buy ratings, six Holds, and two Sells. Argus raised its target to $235 from $180 on Thursday, leaving limited upside from current levels.
Looking ahead, the immediate catalyst is index-related buying, but the longer-term test will be commercial execution. Investors will need to see sustained NovaSeq X demand and margin expansion to justify the premium valuation.
